Holographic imaging and optogenetics of neural circuits

Nervous systems are composed of complex neural circuits, with hundreds of different cell types, synaptically connected in intricate fashion. To explore the function of neural circuits, it would be ideal to use light to record and alter the activity of neurons in three dimensions, while maintaining single cell resolution. To achieve this dream, we have developed holographic microscopy combining two-photon excitation, calcium imaging and optogenetics. I will review our approach and illustrate it with recent data optically altering the behavior of mice, while performing a visual discrimination task.
